GnRH Agonists
Gonadotropin-Release Hormone Agonists (or GnRH agonists) are the safest and most effective class of medication to block Testosterone and other Androgens. The colloquial term "puberty blockers" almost exclusively refers to the GnRH agonists Triptorelin, Goserelin and Leuprorelin, though they are commonly used in adult HRT regimens too. They've been in use around the world since the mid-1980s and study after study has shown them to be both safe and effective.
Due to their method of action GnRH agonists are sex agnostic. They're equally effective for both AMAB and AFAB individuals, and also for most intersex people.
GnRH agonists are typically delivered as a large intramuscular depot injection every 1-3 months, making them impractical for DIY use. However, the larger issue is cost and a lack of reliable supply. When available these medications often cost $300-$1000 per injection.
Recently we've seen cheaper formulations come to market such as Buserelin in a nasal spray and Relugolix in a tablet. For people looking to DIY these are more practical options.
